Indication : Allergic and inflammatory disorders, Rheumatoid arthritis Initial, Multiple sclerosis, Allergic and inflammatory disorders Given as Na⁺ phosphate ester, Conjunctivitis, Otic Allergic and inflammatory conditions of the ear, Intra-articular Joint inflammations

Mechanism of action : - decreases inflammation by inhibition of migration of polymorphonuclear leukocytes and reversal of increased capillary permeability.

                                      - suppresses the immune system by reducing the activity and production of the lymphocytes and eosinophils.

Pregnancy Category C

Contraindication : Hypersensitivity; live vaccines; herpes simplex keratitis, systemic infections.

Precaution : Patients with hypothyroidism, cirrhosis, ulcerative colitis, CHF, convulsive disorders, thrombophloebitis, peptic ulcer, elderly. DM, hypertension, psychological disturbances, osteoporosis.

Drug Interaction : antidiabetic drugs ,isoniazid ,salicylate , Amphotericin B and loop and thiazide

diuretics ,Carbamazepine, phenobarbital , phenytoin , rifampin.

Side Effect : fluid and electrolyte disturbances (with possible edema and hypertension), hyperglycemia

and glycosuria, spread of herpes conjunctivitis, activation of tuberculosis, osteoporosis.

Monitoring parameters : Serum K⁺, BP,glucose ,Observe for  signs or symptoms of Cushing’s syndrome.

Usual Dose : -PO Allergic and inflammatory disorders 5-60 mg/day, in 2-4 divided doses.

      -Rheumatoid arthritis Initial: 5-7.5 mg/day, adjust dose if needed.

      -Multiple sclerosis 200 mg/day for 1 wk, then 80 mg every other day for 1 mth.

       -IV/IM Allergic and inflam dis. Given as Na phosphate ester: 5-60 mg prednisolone base/day.

       -Ophth Conjunctivitis As 0.12 or 1% acetate susp: Instill 1-2 drops 2-4 times/day.

       -Otic Allergic and inflammatory conditions of the ear As 0.5 or 1% acetate or phosphate soln

       -Intra-articular Joint inflam. As acetate: 5-25 mg. As phosphate: 2-30 mg. As terbutate:4-40 mg.

Missed Dose : If you miss a dose or forget to take your medicine, contact your doctor or pharmacist for instructions.

Auxiliary labels  : for PO take w/o food immediately ,for topical applies lightly on  specified location
